Shakib went into the match two wickets short while Mashrafe was just a wicket away at the start of the match at Zohur Ahmed Chowdhury Stadium in Chittagong on Wednesday.
Shakib, however, reached the milestone first with the scalp of Hashim Amla. He became the second Bangladesh bowler to achieve the feat after Abdur Razzak.
Mashrafe joined them later in the match.
Mashrafe, who took the new ball with Mustafizur Rahman on the sluggish pitch, took himself off and introduced Shakib into the attack in the sixth over.
Shakib snared the wicket of in-form Faf du Plessis for his first.
The top allrounder could have had Amla caught in his fourth over to reach the milestone but Sabbir Rahman grassed two attempts at the catch.
Shakib finally sent the Proteas stand-in captain back in his next over when he tossed it up to lure the opener into a drive.
As rain interrupted proceedings, Mashrafe had to wait for some time for his wicket.
Mashrafe’s 200th wicket was crucial as it broke South Africa’s toughest resistance, between David Miller and Jean-Paul Duminy, worth 63 runs.
Miller cut towards point where Sabbir took the catch with a spectacular diving effort.
Left-arm spinner Razzak had achieved the feat in his 141st match in March 2013 when he dismissed Sri Lanka’s Sachithra Senanayake in Pallekele.
